Anyone have a chip/tuner for a 2001 7.3? Also if I buy a used system I have heard that you have to unlock them because the tuner remembers the vin #. Is that true?

It depends.
Programmers/Tuners:
Like HyperTech PowerProgrammer and DiabloSport Predator are married to a truck temporarily. Once you plug it it, you add a Custom Tune to the truck, and the Stock Tune is stored on the programmer. This form of tuning is done via the ODB2 diagnostic port. You tune the truck, and then pull the programmer out and store it in the garage. Usually, you get to make one tune to the truck and leave it that way, until you come home and decide to adjust the tune in your garage/driveway which takes about 20 minutes. If you buy one of these, you have to make sure the Stock Tune has been returned to the truck before you buy it. Otherwise you are hosed. Often, when someone wrecks their vehicle, they sell the used Programmer on eBay, and never return the Stock Tune back to the truck's computer. That's when the buyer gets screwed.
Chips:
Like Bully Dog, SCT, Banks, etc. are not married to a vehicle. They plug in directly to the computer on the truck via an accessory port, AND have to be left in place as long as you want to keep that truck tuned. Usually, they will have some type of switch that you can adjust on the fly between settings. You can easily pull this type of tuner from one truck to another as in to buy used.
Still, buyer beware on any electronics that you buy. That's why most places don't take returns on electronics. They could be hosed, and then you are hosed.

Back from Florida with DP tuner installed but I still have the stock exhaust and air filter, I got a little over a mile per gallon better. I went from just over 15 to between 16 and 17. I hope with the air filter change and exhaust I will get even better. The extra power is pretty awesome. Well worth it.

between 16-17 is what i get, all around town type driving. have not taken trip on highway after install, iam very happy though i was averging between 12.5-13.5 so big increase. i hand calculate numbers.

Just did two back to back trials on my 01 F350 quad cab 4X4. First with stock set up mostly city driving got 16.2 mpg. Second with Hyperchips tuner and installed 60 hp flash. Removed the air filter assembly and installed a big as$ Napa air filter that works like a cold air system but only costs $50 and got 17 7mpg and "WAY" more power and fun to drive. Next a 5" cat back exhaust.
